Story related, can't be missed.
When fighting the final boss, aim for the large eye at the top to deal a critical hit with each successive shot. Keep your distance to avoid its tentacle, but be aware that its shockwave cannot be dodged. If you're a hunter, however, a health-regen class mod will help against this. Best to be done with multiple players on the second playthrough.

An easy way to defeat the Destroyer in second playthrough. Obtain the level 48 MIRV explosives and through all your grenades in the Destroyers mouth. They grenades keep exploding and after that, the Destroyer will only have half a life bar. After that, it's smooth sailing using a Double Anarchy weapon or any other strong weapon or Class Mod.

I found and even easier way (though its cheap). Fire a shot or two from the most powerful weapon you have and run off the edge of the clip either to the left or the right of the entry. You will respawn but the damage done remains. Repeat until dead

@1 I'll have to try that strategy with the MIRVs. I have been using Incendiary Rain grenades, which are great against the Lance and other bandits, but aren't very good against the Vault Boss. What makes it hard (at least for me) is that he levels up with you. I'm level 57 and he's level 59. The last time I fought him I critical hit him with all my sniper and rocket launcher ammo along with a good portion of my combat rifle ammo before I died, only taking out half of his health. So, thanks, I'll give it a shot.

The easiest strategy for this is to have a really good shield and health.
Just make sure all your points are up, i did this on level 58.
so heres what to do, when the boss comes out of the cave and moves the best thing to do is use the guns with unlimited ammo and damage. Then start out by shooting him
run to the right and hide behind a long concrete barriar up standing.
He will start trying to blow you backwards, if you move left and right side to side he cant hit you at all. About half way through the battle his tenticles will come out wrapping around 2 different poles. i chose the right its close the the barrier for cover. i used berserker for this go over and use your special abilty and unlimited guns hit the eye looking thing on the teniticle wrapped around the pole. do this a few

Killed the destroyer (vault boss), who is located in the vault. If you haven't unlocked that area yet, complete more of the main quests to uncover more areas.

If you have Lilith and ammo regeneration capability, here's a strategy that worked for me. Hide behind the left pillar. Step out and unload as many clips as you can of your fastest and most power machine gun. When you start taking damage, use the special action to warp to the the right pillar. Wait for your ability to become available again, then rinse and repeat. The AI doesn't seem to see you move to the other pillar if you warp.

the shock wave CAN be avoided ! ! ! if u stay all the way back near the teleporting station because the ground is lower than the rest of that map, hence the shock wave passing right over your head..

Sooooo......the fabled treasure of the vault is a giant mound of tentacles with a toothy girlypart for a mouth? I can't help but feel a little gyped.
Either way, if you shoot the purple bulges in the middle of the tentacles, they will be blown off. This seems to negate the shockwave and some other abilities. Then just hide behind one of the stone pillars that he grabs and shoot the eye thing between laser blasts. When the laser charges up, hide and reload.
This will empty most of your ammo but with the tentacles gone he can't really hit you so it's pretty easy.

On the second playthrough I sniped the eye and tenticles till he started the shockwaves.
After that he wraps his larger tenticles around the far left and right pillars.
I hid behind the right pillar(one on the left as well) and unloaded my Dove into the larger tenticle.
This is time consuming but the safest way to kill the vault boss solo.
I used the dove simply because you never have to reload and use no ammo.
You could also use any type of ammo regen class or weapon for the same effect.

More or less, the same as #5 by HarrySchnutsack and #12 by Vikairius - did what I could until the larger tenticles came out; ran around looking for cover, found cover on the left behind one big purple tenticle, slapped it a few times, noticed the points floating in the air, slapped it a few more times thinking this could take a very long time, suddenly realised I had weapons(!), and just kept my finger on the trigger until the Destroyer's health wore right down. Went through all my upgraded SMG ammo and finished it off with my sniper. I consider this kinda a cheats way, not really playing the game; but I don't consider myself a hardcore gamer anyway, and I was phase-walking all the way through the Guardians anyway because I just wanted to get to the end.

My strategy for fighting the destroyer was the same for fighting the rakk hive: Have a high pistol proficiency, an sdu of 600 bullets, a gun called cold torment which has fire rate 8.3 and 81 damage, don't use it until you get to the boss, and shoot like hell. Neither the rakk hive or the destroyer had a chance to hurt me before they were dead.

On my first playthrough, I thought getting to the vault was more of a challenge than the boss. I was level 36 when I had killed it and the boss was level 33. Had a machine gun with like 133 damage and my shield was 603. Rarely lost health.
I just threw a few grenades, used up rest of my ammo and sniped it's mouth and got a few criticals. Easy stuff.

Okay I have searched all threads and im guessing this has only happened to me, but right after I killed the destroyer, I lost all my guns and my inventory space went from 21 all the way back to 12.. so during playthrough 2 I have nothing, can anyone explain this?

I thought they woulda made the last boss a little bit more difficult. I agree with previous comments saying its def an anticlimax ending . great game though
