Introduction:
[PROTOTYPE] is an open world sandbox game developed by Radical Entertainment and published by Activision. You play the role of Alex Mercer, a person who wakes up on a morgue slab with no recollection of how he got there, but he has extremely powerful abilities. His goal is to find out who made him the way he is and get his revenge on them.
There are no online modes in [PROTOTYPE], so there are no online achievements. However, you will find yourself getting frustrated while you grind the event achievements attempting to get Platinum medals on them.

So lets start getting those achievements! The first thing you want to do is start a Medium Playthrough.

Step 1 - Medium Playthrough - Set Up:
"Revenge Revisited" requires you to beat the game on Hard difficulty, but before you can even unlock Hard, you must complete the game on Normal, NOT EASY! In this playthrough, you will be getting the majority of the achievements for the game, but also you will be keeping an eye in how you play for two achievements: "Nice Guy" and "Hard To Kill" combined together they score 100 .

For "Nice Guy" you must complete the game while consuming 10 civilians or fewer. To avoid consuming them, try to avoid picking them up with and hitting afterward - that's a consume. It's pretty simple, but also frustrating, to do. "Hard To Kill" requires you to complete the game without dying. Fans of Bioshock will remember the Brass Balls method where whenever you die, quit to the main menu and re-load your last save / auto-save so it doesn't register as a kill. Save often during this playthrough, as you'll need to restart quite often if you're new to the game.

Step 2 - Medium Playthrough:
The only achievement you won't get during this playthrough, should be completing the game on Hard, "Revenge Revisited". Everything else can be achieved in this playthrough.

My advice to get the above to achievements (as mentioned in the set up) easily and quickly is to go straight for the storyline. There are only 31 missions and they shouldn't take too long to finish. When you have those out the way, work towards your other achievements listed in the guide, which include grinding kills on Infected, destroying vehicles, shooting down helicopters, consuming targets, doing events with highest scores possible, etc.

Step 3 - Hard Playthrough:
Rejoice, you can die and you can consume whoever you want. Hard is exactly what it says on the tin, Hard. This won't be an easy ride, but as before, go for the 31 missions and nothing else. After you finish Hard, you can delete the save and go back to your medium playthough so you can:

Step 4 - Mop Up:
No doubt by now you've been playing the game for a while, possibly broken a few controllers and ripped your hair out several times. Grinding for killing the infected will take a while but is not difficult. The actual difficult achievements which give this game an 8/10 for difficulty are the Events. You need to finish them all with Platinum for the "Au" and "Pt" achievements and there are 53.

When going for them, you can find the requirements for Gold and Platinum here.
There are also Collectibles in Prototype. There are three achievements for getting all Hint and Landmark Collectibles, and for consuming all Web Targets. When going for these, I strongly suggest using these maps.

All I can say is good luck, and if there are any you're stuck on, feel free to post in the forums where somebody will be happy to assist you.

x360a would like to thank WhineyRabbit for this Road Map

Revenge Revisited60
Complete Story Mode on hard difficulty. (27)
To unlock the hard difficulty, you must first complete the game on medium difficulty. There are a total of 31 main story missions in the game, so you must complete them all on the hardest difficulty. This will NOT be easy. Hard mode is very tough, I recommend that you remain hidden through as many missions as possible. If spotted, flee and disguise, avoid conflicts with stronger enemies like Super Soldiers and Leader Hunters whenever possible. If you follow the road map, you should be able to die and resume at a checkpoint when you do your hard playthrough.
Endless Hunger10
Consume 200 characters to boost your health. (1)
This will come to you quite quickly. To consume an enemy, you can either do a stealth consume (unlocked with your abilities; hold RB and press B) or use a melee attack to consume anything apart from hunters (grab with B, consume with Y). After any 200 characters (infected, civilians or military) have been consumed, this will unlock.
Trail Of Corpses30
Kill 53,596 infected. (39)
Infected can be any of the Hunter forms and also the infected civilians. If you are trying to boost the achievement, it is better to wait until the city has been approximately 60-80% infected as they will be everywhere. Simply run around killing everything you see as quickly as possible. It will also help if you use infected vision, which will make all infected enemies grow red.

The quickest way to do this is replay the event "Rolling Thunder" (located in the SW corner of the map) which has you using the Thermobolic Tank to kill as many infected as possible. You can easily get 750+ in only a few minutes each time you play this event.
Wrecking Yard20
Destroy 2,000 vehicles. (2)
Vehicles can be cars in the streets, tanks or helicopters. You should get this eventually just by playing through the game as when you're fighting in huge battles with infected and military, you will destroy many normal cars without realizing it. You can attempt this at the same time as Speed Bumps, as you can crush normal vehicles with tanks.
Misconception20
Destroy 25 Infected Water Towers before they hatch. (3)
Near infected zones you will see average looking Water Towers. You are shown these in an early mission. However, when you get close to these towers, they appear infected. You must destroy the tower within 5 seconds of getting too close to it before it hatches and you have a Hunter on your ass. The easiest way that I've found is to use the Whipfist from a distance, so it doesn't hatch, or use the arm blade and slice away quickly. Infected Towers are easily spotted by crows circling them ahead and they are only found in the red zones.
Brain Trust20
Complete all Consume Events. (12)
Consume events appear on your map after a few missions. Complete all of them for this achievement. There are many events for different difficulties. After you complete the first batch of Easy events, you will then unlock Medium, complete these to get Hard events. There are 35 in total.

Easy: 10 Military, 3 Infected. Complete these to unlock Medium.
Medium: 9 Military, 3 Infected. Complete these to unlock Hard.
Hard: 7 Military, 3 Infected. Complete these to unlock the achievement.
Streetwise50
Collect all Landmark collectibles scattered across New York City. (18)
Landmark collectibles are blue orbs scattered across the city. There are 200 of them. You can use this map while collecting them to track your progress.
Polymath40
Collect all Hint Collectibles scattered across New York City. (5)
Hint Collectibles are red orbs scattered around the city. There are 50 of them. You can use this map while collecting them.
Half-Truths10
Complete a mission without causing a single Military Alert. (2)

See "Mankind Is Your Mask"

Mankind Is Your Mask40
Complete 3 missions without causing a single Military Alert.
This can be tricky depending on what missions you attempt to do it on. The first one that I did was when you have to transport the Doctor in the tank. Make sure you don't fire AT ALL, and you won't be discovered. You should also try and avoid running over the military.

The second one would be the mission after this one to protect the doctor. Again, stay disguised the entire time.

Other fairly easy missions are when escorting the Bloodtox Pump; the one after that protecting the Bloodtox Pump; or an earlier mission when you have to escort the Thermobolic Tank.

Important: The stats for this carries over to a New Game + so if you can only get one or two of these done without alert, you can do the same exact mission again on a different playthrough to finish off the achievement.
The Butcher30
Kill 50 characters in 5 seconds. (5)
This is very easy to do. In one of the early missions, you will get to jack an APC tank and, before reaching the Hive you need to destroy, you will have unlimited rocket launcher ammo. Simply go wild on the dashing crowds to obliterate them. You should be able to fire 2-3 rockets in 5 seconds, easily killing 50. Make sure you're also running some over at the same time to help with the kill count.

If you don't get it early on, just wait for the Rolling Thunder event with the Thermobolic Tank. You should have no problem getting it then.
Return Fire10
Catch any object tossed by a Hydra; and throw it back. (3)
All you need to do for this is lock on to a Hydra and wait for it to throw a car. Get ready, and time it so when the car is just about to hit you, press to catch it, throw it back with and, if it hits the Hydra, you'll unlock this.
Gunning10
Destroy 20 Helicopters in a single Helicopter flight. (7)
This is tough. Enemy helicopters can take you down easily with their rockets. I recommend trying this on easy and don't stop moving. Avoid all missiles and fire all missiles you have available at the enemy. When you run out use the guns. The key is to do this on easy and keep moving.

Another method is with a glitch posted by Comatose here.
Cleanup10
Kill 15 characters with a single Whipfist attack. (4)
Use the spinning Whipfist attack, this will hit all enemies around you. Once again, go to Times Square and try on a large group of civilians. To use this attack, equip the Whipfist and hit , . You must first buy this from the Upgrade menu.
The First Thread10
Unlock the Web of Intrigue. (1)

Unlocked around the third mission, cannot be missed.

Self-Deception20
Discover what happened to Alex Mercer through the Web of Intrigue. (3)

Consume James Winninger, you will get this when going for "Web of Knowledge".

Web Of Knowledge60
Acquire all nodes of the Web of Intrigue. (9)

Consume all 130 Intrigue targets.

You can use this map while collecting them to track your progress.

It's Him!10
Patsy 5 military personnel. (2)
Unlock the Patsy technique in the upgrade menu. Then, consume a marine and travel to a base. Walk in casually and use the ability on another marine. You can then either go to a different base, or try on another target. This doesn't have to be done in a base, but it's the best place to do it multiple times. After unlocking Patsy, to use it you need to be hidden, target a Military Personnel with , hold and press .
In Plain Sight10
Evade 10 strike teams.
When causing a Military alert, a Strike Team could be called. When you do get a message appear saying so, start to run away as far as possible. Once you feel you're a safe distance away and the shutter is closed saying you're hidden, swap forms to a civilian and keep running. Once the message "Strike Team Evaded" appears, you've done one. Rinse and repeat.
Infiltrator20
Infiltrate 10 Military Bases disguised as a Commander.
After a certain amount of time, blue circles will appear on your map. In the center of each of these, is a Military Base and each have a Commander nearby. Sneak up on the Commander, consume him and enter the base. Do this with 10 Commanders at their bases.
Threat Elevated20
Destroy 25 strike teams.
Strike Teams are sent after you cause a big military alert. These usually contain three helicopters. Destroy all three to have the Strike Team Destroyed message appear. Rinse and repeat. The easiest way to destroy them is with Whipfist. Either latch onto them and steal them, which will count as destroying them, or use the attack for some serious damage.
Repossession10
Seize 50 vehicles from enemy hands. (2)
This is easy to get when you acquire the whipfist grab. Latch yourself onto a helicopter and hijack them, when you have control of it, you've seized your first. Repeat, or alternatively, you can jack tanks too.
SME20
Gain the ability to drive and fly all vehicles.

Story based and cannot be missed.

Surface-To-Air10
Shoot down 50 helicopters while driving armor. (3)
First of all, do this on easy and be sure to keep moving. In your tank, cause enough fuss to get a strike team to attack, destroy all helicopters and you will have killed at least 3 / 4. This is a cumulative achievement, so you can change tanks whenever you want, or you might get it over the course of the game.
Evolutionary Step10
Use the Upgrade Menu to acquire an Upgrade.
Simple, get some EP, pause the game, go to upgrades, buy whatever tickles your fancy and you will unlock the achievement. You should be able to get this for the first time when you first enter free mode after the third mission.
Unnatural Selection40
Acquire all available upgrades. (7)
This will take quite a bit of grinding. Keep doing missions, events, destroying strike teams etc. and you will eventually earn enough EP to unlock all abilities. If you haven't unlocked it by the end of your first playthrough, start a New Game+ and keep grinding.
The Cleaner20
Destroy 10 Military Bases or Infected Hives in New York City.
You will know when you have found a Hive or Base. The Bases are blue and the Hives are red. The hives are marked as honeycombs and the bases are stars. All you have to do is get either a tank, chopper, rocket launcher and fire away at the building until it falls. The buildings have a health bar, so you will know when it is about to fall. You will acquire this fairly early on if you complete consume events or just like blowing stuff up.
In The Web20
Consume 50 Web Targets.
There are 130 Consume Targets around the city. After 50 have been consumed (not killed), achievement unlocked.
Au40
Achieve a rating of Gold or better in all Events. (1)
There are 53 events in the game scattered around"

10 Movement Events, 10 Glide Events, 24 Kill Events and 9 War Events. When you activate one, you will be given three different targets, bronze, silver and gold. Complete the event with the set target for gold or better to achieve a gold rating. You must get gold on all events for this.

For a list of Gold requirement times for all events, please use this guide.
Nice Guy40
Complete the game while consuming 10 Civilians or fewer. (13)
This is actually surprisingly easy. Play the game going through the missions and just make sure you DO NOT grab civilians. If you do, press to throw them. Do NOT press , or you will consume them. You can keep track of this in your Statistics.
Hard To Kill60
Complete Story Mode in any difficulty without dying. (19)
Bioshock players will know this tactic from Brass Balls. Please note, failing a mission doesn't count as a death, so don't worry about those. Make sure you save your game regularly. If you do die, quit to the main menu and reload your save.
Speed Bumps20
Run over 500 characters in a single tank. (3)
This will take about 10 minutes. In the third or fourth mission, you must rescue your ex-girlfriend. When you first acquire a tank, it will have unlimited ammo and the streets will be very barely infected. Avoid the infected zone and just drive on the sidewalks and run over as many as possible. It's incredibly fun to get this as you see the civilians trying to scatter. Keep going until it pops up.
Pt60
Achieve a Platinum Medal in all Events. (16)
Before you even think about going for Platinum in ANY event, you must first get gold in all 53 and achieve the "Au" achievement. Once you have done so, a notification will appear saying you can now go for Platinum on all events; any event that has already earned a Pt score will be automatically upgraded. Go through all the events again, getting Platinum requirements.

To make some of the events easier (such as Raid and War), finish a playthrough and start a New Game + on Easy to finish them off.

For a list of Platinum requirement times for all events, please use this guide.

Warning: DO NOT get Pt scores on everything before unlocking the Au achievement. For some reason, it appears they do not stack and will glitch, only giving you Au and not Pt, despite all your events being at Pt status. The only way to fix this is to start a new game and do them all over. So, be sure to leave at least one event at a simple Au ranking to unlock the Au achievement and the ability to score platinum, then finish it off with a Pt score if you need. You can do up to 52 at Pt ranking before unlocking the Au achievement.
