Introduction
Welcome to MUA2! This is a great game for playing some co-op with friends, but it can be enjoyed just as well on your lonesome. The first thing you'll need to know is that at this time, the multiplayer aspect is EXTREMELY glitchy and the setup is just plain odd. If you are playing online with friends, you will not earn credit for anything if your save file is further in the game than theirs, nor will you earn anything if your save file is on the opposite side of the war than theirs. So, if you plan to play in co-op AT ALL, make sure you get on the same page as your friends and stay there. Save often as well, and make sure everyone in the game saves before the host leaves!
Oh, and before you begin playing, download this game add-on which is required to connect to other players who have Juggernaut downloaded. Even if you may not play with anyone else, get it to be safe. You'll also need to download the Character & Sim Mission Pack Teaser for the same reasons.
Step 1: Super-Heroic Difficulty
I would suggest starting here over Heroic simply because the achievements "Undefeated Heroes" and "Immortal Heroics" require this to receive credit, not to mention the game is easy as pie anyway. It doesn't really matter which side you take in the war. Do your best not to die at all during this playthrough, though if you do it is not a big deal since you have the mission replay terminals at your disposal. Use the collectibles guides to grab everything as you play, work on the different types of fusions, combat mastery achievements and heroic deeds and also do the five things required for "Seek and Destroy." None of these are critical to focus on too hard since you can use the mission replays and combat simulators to quickly knock out anything you don't get while progressing through the story. However, you will want to unlock any costumes that require this specific side in the war before moving on to your next playthrough. So if you chose "Pro" you'll want to do Iron Man, Mr. Fantastic, Songbird, etc. Check that achievement for more info.
Step 2: Legendary Difficulty
Once you finish Super-Heroic, you will unlock this difficulty. All your stats will carry over with you, so start a new game by using the "next mission" console in the Wakanda HQ and be sure to choose the opposite side in the war as you did last time at the end of Act 1. Again, use the collectibles guides to grab anything you may have missed before, plus you will have all-new levels during Act 2 to focus on. Now once you finish playing the game again, you should have all story progression achievements and all collectibles from the missions.
Step 3: Mop Up
You can use the mission replay terminal to now mop up anything you've missed. Grab a friend and go through all the Trivia questions to get total bonuses, go through all the combat simulations to get Gold ratings, repeat HQ levels to get 24 co-op maps, finish off any Undefeated levels and/or bosses you've missed and knock out any heroic deeds you still need for your final team boosts.
Step 4: Character & Sim Mission Pack
Unfortunately with the DLC no longer available except for the "teaser" (which allows you to play online with those who do have it), you won't be able to get the full 1250 if don't already have the DLC. For those that do have the DLC, there are achievements that can be done in one run ("Low Deductible" and "Hotfoot") and some lower difficulty achievements (such as "Old Foes") that keep this DLC pretty short. Roughly 2-3 hours should be plenty to finish off the full 250

Conclusion
Ultimately (no pun intended) Marvel Ultimate Alliance 2 is a worthy sequel to the original and can provide hours of entertainment and multiple playthroughs.
